There are a few ways to get to the 33 but I think the quickest way is taking the Ojai33 exit off the 101. The 33 is a nice detour if you want to take the scenic route from SoCal north to the Central coast and only adds an hour or so to the trip from Los Angeles to San Luis Obispo.

Running from one side of the 33 over the hill to the other should take anywhere from 30 minutes to an hour depending on how quick you are. The first half can be pretty calm on a windy day and then winds suddenly pick up once you crest the hill and start coming down the north side so watch out for that.

There is a ranger station that makes a good resting place / turn around point if you don't intend to continue up to the 166. It's right before the dead straight away that leads to the 166.
